The nerve growth factor/tumor necrosis factor receptor family.

چکیده
Receptors in the nerve growth factor/tumor necrosis factor receptor family are characterized by the presence of cysteine-rich motifs of approximately 40 amino acids in the extracellular domain. The ligands are type II transmembrane proteins with beta-strands that form a jelly-roll beta-sandwich. The receptors recognize soluble or cell-surface-bound ligands and mediate diverse cellular responses. Activation of intracellular signals is mediated at least in part by the association of proteins with a RING finger motif or a death domain to the cytoplasmic domains of the receptors. In addition to cell-membrane-bound receptors soluble forms have been described for most of the receptors. Activation of intracellular signals not only occurs through ligand binding to the receptors but cross-linking of at least some members of the ligand family can regulate cell functions.

INTRODUCTION The tumor necrosis factor receptor (TNFR) family consists of a number of type I transmembrane glycoproteins characterized by homologous cysteine-rich domains in their extracellular region. The intracellular parts of these proteins vary in size and structure, corresponding to the wide array of functions of TNFR proteins, ranging from regulation of cell activation and differentiation...
متن کاملAssociation of VNTR polymorphism of tumor necrosis factor receptor 2 (TNFRSF1B) with pulmonary tuberculosis
This study was designed to find out the impact of the variable number of tandem repeats (VNTR) of the tumor necrosis factor receptor 2 (TNFRSF1B) on pulmonary tuberculosis (PTB) risk in an Iranian population. This case-control study was done on 159 PTB patients and 158 healthy subjects. Bi-allelic TNFRSF1B VNTR was genotyped by polymerase chain reaction. Logistic regression analysis revealed no...
متن کاملA new member of the tumor necrosis factor/nerve growth factor receptor family inhibits T cell receptor-induced apoptosis.
By comparing untreated and dexamethasone-treated murine T cell hybridoma (3DO) cells by the differential display technique, we have cloned a new gene, GITR (glucocorticoid-induced tumor necrosis factor receptor family-related gene) encoding a new member of the tumor necrosis factor/nerve growth factor receptor family. GITR is a 228-amino acids type I transmembrane protein characterized by three...
متن کاملCD 271 (P75 neurotrophin receptor).
P75NTR (or CD271) is a member of the Tumor Necrosis Factor receptor (TNFR) super family of transmembrane proteins that share significant homology in their extracellular domains. Subsets of TNF receptors, including CD271, have a cytoplasmic death domain, although CD271 has unique intracellular structure and downstream signaling partners. CD271 is also differentiated from other members of the TNF...
متن کاملCharacterization of a p75(NTR) apoptotic signaling pathway using a novel cellular model.
The p75 neurotrophin receptor (p75(NTR)) belongs to the tumor necrosis factor receptor/nerve growth factor receptor superfamily. In some cells derived from neuronal tissues it causes cell death through a poorly characterized pathway. We developed a neuronal system using conditionally immortalized striatal neurons, in which the expression of p75(NTR) is inducibly controlled by the ecdysone recep...
